This assay is designed to measure phospholipid fatty acids in plasma, breast milk, food or tissues. The following phospholipid fatty acids can be measured by GC-FID:

All samples are extracted once and injected twice (see the SOP) for details).

Please note that some phospholipid fatty acids in serum or plasma are represented at very low levels that are at, or near, the limit of detection by the GC-FID method. As such, the %cv for these is significantly higher than that for the more highly represented fatty acids.

 

Sample Collection Instructions

Plasma or Serum: Blood should be collected into a 4 mL EDTA or 6 mL heparin tube, inverted and placed on ice. Red cells are removed by centrifugation for 10 minutes at 2000 x g at 4°C. Plasma is collected and stored as 500 µL aliquots in 1.5 mL tubes at -80°C until ready to ship. Ship on dry ice for overnight delivery. Serum can be collected using SST-5 mL tubes. For accurate results, it is imperative you send the specified volume. Accurate analyses are not possible with less than 500 µL.

Breast Milk: Collect as much milk as possible from one breast in clean container. Please collect all the milk prior to aliquoting as there are differences in foremilk and hindmilk. Cap and mix well, and store 300 µL aliquots in 1.5 mL tubes at -80°C until ready to ship. If collection occurs away from the lab, short term storage at -20°C is okay. Ship samples on plenty of dry ice for overnight delivery.

Urine: Collect urine using a ‘clean catch’ technique into a clean container. Cap and mix well. If it is a 24hr collection, record total volume and period of collection. Store 300 µL aliquots in 1.5 mL tubes at -80°C until ready to ship. Ship samples on plenty of dry ice for overnight delivery.

Food: If sample is a solid food, homogenize with a blender if necessary. Aliquot 1 gram liquefied food into 1.5 mL tubes and store at -80°C. Ship samples on plenty of dry ice for overnight delivery.

Tissue: Collect tissue by freeze-clamping in liquid nitrogen with pre-cooled freeze-clamping tongs. Pulverize frozen tissue under liquid nitrogen with mortar and pestle. Be sure to pre-cool the mortar and pestle and add liquid nitrogen to the mortar before adding tissue. Grind tissue to a fine powder (no chunks) in continuous presence of liquid nitrogen. Transfer powder to pre-cooled tubes. Next, weigh at least 500 mg pulverized tissue into 1.5 mL tubes and store at -80°C. Ship samples on plenty of dry ice for overnight delivery. Please note that pulverized tissue samples must be carefully weighed.

 

Important Notes

  • Send the amount of sample requested and be sure to specify weights and volumes!
  • Sample tubes must be numbered clearly and simply, e.g. 1, 2, 3,…100, and accompanied by a sample key (which also must be emailed) describing the sample, sample volume or weight, and position in sample box.
  • Ship samples overnight delivery with enough dry ice to last 72 hours in case the package is delayed. It happens all too frequently.
  • If shipping a cell pellet, do not forget to determine the cell number, cell protein, or cellular DNA for data normalization prior to shipping. We do not do not perform those analyses.
